[Abstract]
Objective:Through detecting the infection rate of HPV16/18 and the expression rate of MSH2 protein,we can investigate the relationship with HPV infection in colorectal cancer,and study the correlation between HPV infection and MSH2 protein expression,then approach the etiological factor and pathogenesis of colorectal cancer.Method:We detected he infection of HPV 16/18 and the expression of MSH2 protein using in situ hybridization(ISH)and immunehistochemical(IHC).Result:1.the HPV16/18 infection rate of normal mucous membrane of colorectum, colorectal adenoma,colorectal cancer.The HPV16/18 infection rate is increasing progressively in the three tissue,and the difference has statistical significance(P?0.05).the infection rate of colorectal cancer is obviously higher than that of colorectal adenoma and normal colorectal tissue,and the infection rate of colorectal adenoma is higher than that of normal colorectal tissue,the differences both have statistical significance(P?0.05).2.HPV16/18 infection of colorectal cancer is related to its tissue differentiation,Duke?s staging and lymph nodes metastasis,the HPV16/18 infection of poorly differentiated adenocarcinoma is higher than that of moderately differentiated adenocarcinoma and well differentiated adenocarcinoma (the different has statistical significance,P?0.05);the HPV16/18 infection of Duke?s staging C and D colorectal carcinoma tissue is higher than Duke?s staging A and B carcinoma tissue(the different has statistical significance,P?0.05);the HPV16/18 infection of lymph nodes metastasis group is higher than no lymph nodes metastasis group(the different has statistical significance,P?0.05);and is not related to its age,gender,tumor size,locus, and tissue typing.3.the expression rate of MSH2 in normal mucous membrane of colorectum, colorectal adenoma,colorectal cancer.The expression rate is decreasing progressively in the three tissue,and the difference has statistical significance(P?0.05).the expresion rate of colorectal cancer is lower than that of colorectal adenoma and normal colorectal tissue,and the difference between colorectal cancer and adenoma and para-cancer normal tissue has statistical significance(P?0.05).4.the expression rate of MSH2 protein in colorectal cancer is related to its tissue differentiation,Duke?s staging and locus,the MSH2 protein expression of well differentiated adenocarcinoma is higher than that of moderately differentiated adenocarcinoma and poorly differentiated adenocarcinoma(the different has statistical significance,P?0.05);the MSH2 protein expression of Duke?s staging A and B colorectal carcinoma is higher than that of Duke?s staging C and D(the different has statistical significance,P?0.05);the MSH2 protein expression of left hemicolon and rectum is higher than right hemicolon (the different has statistical significance,P?0.05);and is not related to its age, gender,tumor size,and tissue typing. 5.the relationship between HPV16/18 infection and MSH2 protein expression is a negative correlation in colorectal cancer.Conclusion:1.Both colorectal cancer and adenoma have HPV16/18 infection,and the infection rate of cancer is higher than that of adenoma,but no infection was detected in normal tissue,these suggest that HPV16/18 infection plays a promotive effect on colorectal tumorigenesis.2.Both colorectal cancer and adenoma have missing of MSH2 protein,and the miss rate of cancer is higher than that of adenoma,but MSH2 protein is high expression in normal tissue,these suggest that MSH2 protein loss exists in colorectal neoplasm,and may play an important effect on tumorigenesis.3.the relationship between HPV infection and MSH2 protein expression is negative correlation,suggested that the two may cooperate in colorectal tumorigenesis.
